Lope de Zayas y Garcia de Eslava was born in Écija, Spain, in about 1434. He married Maria Mendez de Biedma de Sotomayor in Córdoba, and they had five children, Luis de Biedma, and Gaspar, Francisco, Lope, and Luis de Zayas y Méndez de Biedma. Luis is elsewhere called Luis de Zayas y Méndez de Sotomayor.

Lope was regidor [councilman] de Écija, alcalde [mayor] de Antequera, and one of the best lancers in his time, in Andalusia. He was murdered by the family and friends of Don Carlos de Torres, one of the most powerful families in the town of Jaén, following the elopement of Torres' daughter and the brother of Lope, Gonzalo de Zayas.

Lope de Zayas was poisoned while attending the wedding feast, in Baena, of one Luis Portocarrero and the daughter of the Count of Cabra. Information found in the records of Écija and Granada say that he was placed inside a hanging iron cage by Portocarrero, publicly humiliated, and poisoned.

He was, in either case, buried in the family chapel in Écija.
